Dr. Amol Kolhe was born in Narayangaon near Pune. He studied up to class VIII there and then came to Pune for further education. He studied up to 12th standard in science from Apte School. Seth Govardhandas Sundardas (G.S.) From college Dr. Amol Kolhe completed his medical education in MBBS.

Dr. Amol Kolhe already considers Chhatrapati Sambhaji Maharaj as his role model. Dr. Amol Kolhe joined the Nationalist Congress Party in March 2019 and was elected as an MP. In 2024, he again contested the Shirur Lok Sabha elections and won.
